A half-life extended monoclonal antibody targeting kallikreins 5 and 7, in Phase 2 proof-of-concept trial for atopic dermatitis.
TRIV-509 is a half-life extended monoclonal antibody that selectively targets kallikreins 5 and 7 (KLK5/7), key enzymes implicated in skin barrier dysfunction, inflammation, and itch. Preclinical data demonstrated superior efficacy compared to IL-4R inhibition in atopic dermatitis models, and human explant studies showed improved barrier integrity, reduced epidermal thickness, and elevated levels of a key protein needed for skin barrier function.
The first patient has been dosed in a global Phase 2 proof-of-concept trial for moderate-to-severe atopic dermatitis. TRIV-509 is positioned for quarterly subcutaneous dosing and is designed to address the root cause of the disease rather than merely modulating inflammatory pathways.
