Atirmociclib is a next-generation, orally bioavailable small molecule that selectively inhibits cyclin-dependent kinase 4 (CDK4) while sparing CDK6. By narrowing activity to CDK4, the candidate is designed to preserve the hematopoietic role of CDK6 and thereby lessen the severe neutropenia associated with earlier pan-CDK4/6 therapies, while still disrupting the G1-to-S cell-cycle transition that drives HR-positive breast tumor growth.
